TCF-4 Antibody (F-7) is a mouse monoclonal IgG3 antibody that detects TCF-4 in mouse, rat, and human samples through applications such as western blotting (WB), immunoprecipitation (IP), immunofluorescence (IF), and enzyme-linked immunosorbent assay (ELISA). TCF-4, a member of the T cell factor family, plays a crucial role in lymphoid cell development and is a key transcriptional activator in the Wnt signaling pathway. TCF-4 is primarily located in the cytoplasm, where TCF-4 forms a complex with β-catenin before translocating to the nucleus, a process that is vital for the regulation of Wnt target genes, many of which are associated with cancer. The interaction between TCF-4 and β-catenin is essential for the transcriptional activation of these genes, and the presence of the high mobility group (HMG) 1 box motif in TCF-4 facilitates DNA binding. Additionally, TCF-4′s interaction with c-Jun, which is modulated by phosphorylation, highlights TCF-4′s role in integrating various signaling pathways that influence intestinal tumorigenesis. Beyond implications in cancer, TCF-4 has also been linked to bipolar affective disorder, underscoring TCF-4′s importance in both developmental and pathological processes.
