PSGL-1 Antibody (H-1) is a mouse monoclonal IgG3 antibody that detects PSGL-1 in human samples through applications such as western blotting (WB), immunoprecipitation (IP), immunofluorescence (IF), and enzyme-linked immunosorbent assay (ELISA). PSGL-1, also known as P-Selectin glycoprotein ligand or CD162, is a type 1 membrane protein that plays a crucial role in the immune response by mediating the adhesion of leukocytes to endothelial cells during inflammation. PSGL-1 is primarily located on the tips of microvilli of leukocytes, where a unique structure, characterized by a rich composition of serines, threonines, and prolines, allows for effective interaction with selectins. The presence of a series of decameric repeats in PSGL-1′s extracellular domain enhances binding capabilities, particularly with P-Selectin and E-Selectin, which are essential for leukocyte trafficking. For optimal selectin engagement, PSGL-1 must undergo specific post-translational modifications, including sialylation and fucosylation, which are critical for function in mediating cell adhesion and signaling.
