PCID2 (PCI domain containing 2) encodes a PCI-domain protein implicated in the assembly and regulation of large multiprotein complexes that coordinate RNA metabolism and gene expression control. PCID2 has been linked to post-transcriptional regulatory processes, including mRNA surveillance and turnover, and is studied in the context of proteostasis-associated pathways that shape cellular homeostasis. Perturbation of these processes can influence cell-cycle progression, stress responses, and differentiation programs by altering transcript stability and protein output. Dysregulated RNA processing and quality-control networks are frequently associated with oncogenic states and other complex diseases, making PCID2 a useful target for mechanistic studies of gene regulatory circuitry.
PCID2 CRISPR/Cas9 KO Plasmid (h) is a pool of plasmids designed for targeted disruption of the PCID2 gene in human cell lines. Each plasmid co-expresses a unique single guide RNA (sgRNA) targeting a distinct site within the PCID2 together with the Streptococcus pyogenes Cas9 nuclease. The plasmids also encode GFP, allowing fluorescent identification and enrichment of successfully transfected cells by fluorescence microscopy or flow cytometry.
The multi-guide design increases the likelihood of generating insertions or deletions (indels) that disrupt the PCID2 open reading frame following Cas9-mediated double-strand break formation. DNA breaks introduced by the CRISPR/Cas9 system are repaired through endogenous non-homologous end joining (NHEJ) pathways, frequently resulting in frameshift mutations that abolish PCID2 protein expression.
This CRISPR knockout system enables efficient generation of PCID2-deficient cell models for investigation of PCID2 signaling, functional genomics studies, cancer biology research, and evaluation of therapeutic responses in human cell lines.
