I couldn't get any good immunoblot even after loading 80 ug of protein on a 8% SDS PAGE with Biorad nitrocellulose membrane. Very faint or ghost band are visible. I am using RIPA buffer for lysis and I work on MSCs. Any help will be greatly appreciated.

Thank you for your question. We have not tested this antibody on MSC's, and do not know how abundant MRP1 is in that sample. We recommend using T98G, A549 or AML-193 whole cell lysates as positive controls with our MRP1 antibodies. We recommend loading 50 µg protein, and using the primary antibody at a dilution of 1:200. We would highly recommend a replacement mouse monoclonal, MRP1 Antibody (QCRL-1): sc-18835 (https://www.scbt.com/scbt/product/mrp1-antibody-qcrl-1).
