LDH Antibody (H-10) is a mouse monoclonal IgG2a kappa light chain antibody that detects lactate dehydrogenase (LDH) protein of mouse, rat, and human origin by western blotting (WB), immunoprecipitation (IP), immunofluorescence (IF), immunohistochemistry, and enzyme-linked immunosorbent assay (ELISA). LDH (H-10) antibody is available in both non-conjugated and various conjugated forms, including agarose, horseradish peroxidase (HRP), phycoerythrin (PE), fluorescein isothiocyanate (FITC), and multiple Alexa Fluor® conjugates. The lactate dehydrogenase family, including LDH-A, LDH-B, and LDH-C, plays a crucial role in cellular metabolism by catalyzing the conversion of L-lactate and NAD to pyruvate and NADH, marking the final step of anaerobic glycolysis. This enzymatic activity is vital for energy production, especially in tissues that rely heavily on anaerobic metabolism, such as muscle during intense exercise. Each LDH isoform exhibits distinct tissue distribution, with LDH-A predominantly found in muscle, LDH-B in the heart, and LDH-C restricted to the testis and sperm, making these isoforms important markers for various physiological and pathological conditions, including germ cell tumors. The genes encoding human LDH-A and LDH-C are located on chromosome 11, while the LDH-B gene is found on chromosome 12, and deficiencies in the LDH-A gene have been associated with exertional myoglobinuria, highlighting the clinical significance of LDH in metabolic disorders.

I used this antibody on frozen brain sections. The protein should be broadly expressed in the cortex but the staining is not showing any positive neuron, Can you please help me? I used 15 and 40 microm slices using antigen retrieval and permeabilization.

If I want to make sandwich immunoassay for human LDH using this antibody (H-10) as capturing (or detection), what antibody should I use as a counter part? Can you suggest antibody pairs for LDH sandwich assay?

Thank you for your question. Unfortunately we do not support sandwich ELISA and have not tested these antibodies for that use. Any LDH antibody with a different epitope should be able to work for you, so sc-137243 might be a good choice as it is made to a different section of the protein.

Will this antibody (H-10, sc-133123) detect bovine LDH-B in a heart extract on WB analysis?

Thank you for your question. This antibody hasn’t been tested in bovine samples. This monoclonal antibody was raised against amino acids 173-332 mapping at the C-terminus of LDH-A of human origin. This antigen shares 72% sequence identity with LDHB in Bos taurus (NP_776525) [and 93% with LDHA in bovine]. Based on this sequence analysis, the antibody may cross-react with these proteins in bovine samples.

Can you recommend a positive control to use with LDH (H-10): sc-133123 monoclonal antibody for Western blot?

We recommend MDA-MB-231 (sc-2232), Ca Ski (sc-364360) or ES-2 (sc-24674) whole cell lysates to use as positive controls as we have Western blot data showing specific detection of LDH in these cell lines.

Rated 1 out of 5 by from Doesn't work in Leukemia CellsWe tested this antibody in two different Leukemia cells (K-562 and U-937) using the following protocol: Blocking in 5% no-fat milk for 1 hour. Anti-LDH (H-10) 1:500 Overnight. Goat anti-Mouse 1:2000 at room temperature for 1 hour. Sadly, no bands have been detected.

Il segnale risultava molto forte perciò ho effettuato diluizioni intermedie fino ad arrivare anche a 1:5000, la quale ritengo sia quella più appropriata per apprezzare delle differenze.
