KCTD3 CRISPR/Cas9 KO Plasmid (h) | sc-412124 | 20 µg | $397.00 |
KCTD3 (potassium channel tetramerization domain containing 3) encodes a BTB/POZ domain–containing protein implicated in protein–protein interactions that can shape the assembly of multiprotein complexes and influence proteostasis. Members of the KCTD family are frequently linked to cullin-3 (CUL3)–associated ubiquitin ligase biology and to the regulation of signaling intermediates through targeted turnover, suggesting roles in pathway tuning and cellular homeostasis. Through these interaction networks, KCTD3 is relevant to studies of cell growth control, stress responses, and differentiation-associated programs where ubiquitination-dependent remodeling is prominent. Dysregulation of BTB/KCTD-centered complexes has been associated with disease-relevant phenotypes in cancer and neurobiology, supporting investigation of KCTD3 in mechanistic models of altered signaling and protein quality control.
KCTD3 CRISPR/Cas9 KO Plasmid (h) is a pool of plasmids designed for targeted disruption of the KCTD3 gene in human cell lines. Each plasmid co-expresses a unique single guide RNA (sgRNA) targeting a distinct site within the KCTD3 together with the Streptococcus pyogenes Cas9 nuclease. The plasmids also encode GFP, allowing fluorescent identification and enrichment of successfully transfected cells by fluorescence microscopy or flow cytometry.
The multi-guide design increases the likelihood of generating insertions or deletions (indels) that disrupt the KCTD3 open reading frame following Cas9-mediated double-strand break formation. DNA breaks introduced by the CRISPR/Cas9 system are repaired through endogenous non-homologous end joining (NHEJ) pathways, frequently resulting in frameshift mutations that abolish KCTD3 protein expression.
This CRISPR knockout system enables efficient generation of KCTD3-deficient cell models for investigation of KCTD3 signaling, functional genomics studies, cancer biology research, and evaluation of therapeutic responses in human cell lines.
