ILBP Antibody (4A4) is a mouse monoclonal IgG2a antibody that detects ileal lipid binding protein (ILBP) in human samples through applications such as western blotting (WB), immunoprecipitation (IP), and enzyme-linked immunosorbent assay (ELISA). ILBP plays a crucial role in lipid metabolism by binding and transporting both bile acids and long-chain fatty acids, which is essential for the active uptake of bile acids in the ileum. This process is vital for the solubilization and transport of dietary lipids from the liver, ensuring that nutrients are effectively absorbed in the digestive system. Structurally, ILBP is characterized by ten antiparallel beta strands arranged in two nearly orthogonal beta sheets, resembling a clam shell, and is further stabilized by two short, nearly parallel alpha helices. The binding of fatty acids or bile acids to ILBP induces conformational changes that enhance ILBP′s affinity for bile acids, creating a positive-feedback regulation mechanism that is critical for maintaining lipid homeostasis. The human ILBP gene is located at position 5q35 and is predominantly expressed in the small intestine, highlighting ILBP′s importance in gastrointestinal physiology and metabolic regulation.
