FFAR2 encodes GPR43, a short-chain fatty acid (SCFA)–sensing G protein-coupled receptor activated by acetate, propionate, and butyrate produced by the gut microbiota. GPR43 couples primarily to Gαi/o and Gαq pathways to modulate cAMP signaling, intracellular calcium flux, and downstream MAPK/ERK responses, shaping chemotaxis, cytokine production, and barrier-associated immune regulation. It is prominently expressed in immune cells such as neutrophils and macrophages and in metabolic tissues, linking microbial metabolites to inflammatory and energy homeostasis programs. Dysregulated FFAR2/GPR43 signaling has been associated with inflammatory disorders and metabolic phenotypes, supporting mechanistic studies of host–microbiome signaling in disease-relevant contexts.
GPR43 CRISPR/Cas9 KO Plasmid (h) is a pool of plasmids designed for targeted disruption of the FFAR2 gene in human cell lines. Each plasmid co-expresses a unique single guide RNA (sgRNA) targeting a distinct site within the FFAR2 together with the Streptococcus pyogenes Cas9 nuclease. The plasmids also encode GFP, allowing fluorescent identification and enrichment of successfully transfected cells by fluorescence microscopy or flow cytometry.
The multi-guide design increases the likelihood of generating insertions or deletions (indels) that disrupt the FFAR2 open reading frame following Cas9-mediated double-strand break formation. DNA breaks introduced by the CRISPR/Cas9 system are repaired through endogenous non-homologous end joining (NHEJ) pathways, frequently resulting in frameshift mutations that abolish GPR43 protein expression.
This CRISPR knockout system enables efficient generation of FFAR2-deficient cell models for investigation of GPR43 signaling, functional genomics studies, cancer biology research, and evaluation of therapeutic responses in human cell lines.
