CH 223191 is shown to be a potent 2,3,7,8-Tetrachlorodibenzo-p-dioxin (TCDD)-induced aryl hydrocarbon receptor inhibitor. Mechanistic experiments show that CH 223191 blocks binding of TCDD to AhR and prevents TCDD-mediated nuclear translocation and AhR DNA binding. Detailed studies show that CH 223191 specifically prevents expression of cytochrome P450 enzyme, which is the target gene of AhR. AhR is a transcription factor that mediates cell cycle progression and affects transforming growth factor-beta (TGF-beta) signaling.
