B7RP-1 Antibody (HK5.3) is a rat monoclonal IgG2a antibody that detects B7RP-1 protein of mouse origin by flow cytometry (FCM). Anti-B7RP-1 antibody (HK5.3) is available as a non-conjugated format, specifically designed to facilitate the study of T cell costimulatory mechanisms. B7RP-1, also known as inducible costimulator (ICOS)/B7-related protein-1 (B7RP-1, B7-H2, GL50, ICOS-L), plays a crucial role in immune responses by serving as a ligand for the ICOS receptor, which is essential for initiating T and B cell proliferation and cytokine secretion. This interaction is particularly important in peripheral lymphoid organs such as the spleen and lymph nodes, where B7RP-1 is involved in T cell-dependent B cell activation. Notably, B7RP-1 is expressed in myeloid leukocytes, and B7RP-1 transcripts can be detected in various tissues, including brain, heart, kidney, and liver, with lower levels in colon and thymus. B7RP-1 expression is significantly enhanced by pro-inflammatory cytokines such as tumor necrosis factor α (TNFα), granulocyte-macrophage colony-stimulating factor (GM-CSF), and interleukin 4 (IL-4). Furthermore, lipopolysaccharide (LPS) induces B7RP-1 upregulation through a MyD88-dependent signaling pathway, highlighting B7RP-1′s role in the immune response and potential implications in therapeutic strategies targeting immune modulation.
