ACE Antibody (H-9) is a mouse monoclonal IgA kappa light chain antibody engineered to detect angiotensin-converting enzyme (ACE), also known as CD143 or Angiotensin Converting Enzyme 1. Anti-ACE antibody (H-9) is designed to recognize a precise epitope between amino acids 1279-1306 at the C-terminus of human ACE, ensuring accurate detection of both somatic and testis isoforms of ACE from mouse, rat, and human origins through applications such as western blotting (WB), immunoprecipitation (IP), immunofluorescence (IF), and enzyme-linked immunosorbent assay (ELISA). ACE monoclonal antibody (H-9) is an essential tool for researchers studying the structural and functional aspects of ACE, a zinc metallopeptidase pivotal in the renin-angiotensin system. The structure of ACE consists of two homologous domains, each containing an active site critical for enzymatic activity in converting Angiotensin I to the potent vasopressor hormone Angiotensin II, which plays a key role in regulating blood pressure and maintaining electrolyte balance. Post-translational modifications, particularly N-linked glycosylation, are essential for proper folding, stability, and enzymatic function of ACE, facilitating interaction with substrates and inhibitors. These modifications are crucial for ACE′s role in inactivating bradykinin, a vasodepressor peptide, thereby contributing to fluid and electrolyte homeostasis. Two main isoforms exist: pulmonary ACE (ACEP), a glycoprotein expressed in vascular endothelial cells of the lung, liver, adrenal cortex, pancreas, kidney, and spleen, and testicular ACE (ACET), exclusively expressed in adult testis by developing sperm cells, specifically late pachytene spermatocytes. Understanding ACE′s post-translational modifications and structural integrity is vital for researching cardiovascular diseases, hypertension, and related disorders, making ACE (H-9) antibody an indispensable reagent in biomedical research.
