1-Methyl-D-tryptophan is an inhibitor of the enzymes IDO and IDO2 (indolamine 2,3-dioxygenase 1 and 2) with selectivity for IDO2. The enzymes perform similar transformations and are responsible for catalyzing the rate-limiting step of oxidative tryptophan catabolism in the kynurenine pathway. IDO activity is correlated with an induction of tolerance and immunosuppresion through activation of regulatory T cells by metabolites generated from tryptophan catabolism. Inhibition of IDO by 1-Methyl-D-tryptophan blocks this induced immune suppression, which has shown utility in suppressing acquired immunities of tumors and indicates potential for chemical intervention of chronic inflammatory diseases.
