    Overview

    Tpm2 encodes tropomyosin β, an actin-binding coiled-coil protein that polymerizes along F-actin to stabilize thin filaments and regulate access of myosin motors and actin-associated factors. In muscle and non-muscle cells, tropomyosin isoforms contribute to contractility, cytoskeletal tension, cell shape, and mechanotransduction by tuning actin filament dynamics within stress fibers and sarcomeric structures. Through its role in actomyosin organization, TPM2 influences processes such as cell migration, adhesion remodeling, and force generation that interface with Rho GTPase–dependent cytoskeletal pathways. Dysregulation of tropomyosin composition and actin filament control is relevant to disorders of muscle structure and function and to cytoskeleton-driven phenotypes studied in developmental and disease models.

    Tropomyosin β CRISPR/Cas9 KO Plasmid (m) is a pool of plasmids designed for targeted disruption of the Tpm2 gene in mouse cell lines. Each plasmid co-expresses a unique single guide RNA (sgRNA) targeting a distinct site within the Tpm2 together with the Streptococcus pyogenes Cas9 nuclease. The plasmids also encode GFP, allowing fluorescent identification and enrichment of successfully transfected cells by fluorescence microscopy or flow cytometry.

    The multi-guide design increases the likelihood of generating insertions or deletions (indels) that disrupt the Tpm2 open reading frame following Cas9-mediated double-strand break formation. DNA breaks introduced by the CRISPR/Cas9 system are repaired through endogenous non-homologous end joining (NHEJ) pathways, frequently resulting in frameshift mutations that abolish Tropomyosin β protein expression.

    This CRISPR knockout system enables efficient generation of Tpm2-deficient cell models for investigation of Tropomyosin β signaling, functional genomics studies, cancer biology research, and evaluation of therapeutic responses in human cell lines.
