Sp1 항체(E-3)는 WB, IP, IF 및 ELISA에 의해 마우스, 랫트 및 인간 출처의 Sp1을 검출하는 IgG2a 마우스 단일클론 Sp1 항체이다. Sp1 항체(E-3)는 비 conjug 결합 항-Sp1 항체 형태뿐만 아니라 아가로오스, HRP, PE, FITC 및 다중 Alexa Fluor® 접합체를 포함한 다중 접합 형태의 항-Sp1 항체로도 사용할 수 있다. Sp1은 GGGCGGCGGC 및 밀접하게 관련된 서열을 인식하는 서열 특이적 전사 인자로, 종종 GC 박스라고 불린다. Sp1은 처음에 SV40 촉진체로부터 시험관 내 전사를 선택적으로 활성화하고 SV40의 21-bp 반복 요소에서 다중 GC 박스에 결합하는 HeLa 세포 유래 인자로 확인되었다. DNA 결합의 서열 특이성은 Zn(II) 손가락에 의해 부여되는 반면, Sp1의 다른 영역은 DNA 결합의 친화성을 조절하는 것으로 보인다. Sp1은 촉진체 서열에 결합할 때 인산화되는 전사 인자의 하위 그룹에 속한다. 증거는 초기 성장 반응 유전자인 Erg-1(Zif268 또는 NGF1-A로도 알려져 있음)이 Sp1과 중복 결합 모티프에 대한 결합을 위해 경쟁함으로써 특정 포유동물 유전자 촉진체를 하향 조절할 수 있음을 시사한다. 인간 Sp1을 암호화하는 유전자는 염색체 12q13.1에 매핑된다.

Does it have any cross reactivity? Can I use it in Yeast?

This antibody is currently recommended for detection of Sp1 in mouse, rat and human samples. Please contact our Technical Service Department and we would be happy to perform sequence analysis using the antigen to predict any potential cross-reactivity with yeast. The antibody has not been previously published in yeast.

Can the Sp1 Antibody (E-3) X (recommended for ChIP applications) also be used in regular IP applications?

Thank you for your question. Yes, the "X" product, or TransCruz reagent for ChIP applications (sc-17824 X) of our mouse monoclonal, Sp1 Antibody (E-3): sc-17824, is suitable for regular immunoprecipitation (IP) applications. The only difference is that the antibody is provided 10 times more concentrated, at 200 µg/0.1 ml, and the buffer does not contain gelatin.

I am using the LiCor/Odyssey system. Which secondary antibody do you recommend using with Sp1 Antibody (E-3): sc-17824?

We recommend using one of our our exclusive Mouse IgG Binding Proteins (m-IgGκ) conjugated to CFL 680 or CFL 790. They are suitable for use with Near-Infrared (NIR) detection systems, such as LI-COR/Odyssey and other comparable systems. Rated 4 out of 5 by from Great Results on Western BlotI got very strong and specific reactivity on Western Blot with 50 ug of HeLa Lysate at 1:250 dilution. I only did that much because I was testing it but I will keep using this antibody and will be loading a lot less protein and diluting the antibody a lot more.
