Mouse Sct encodes secretin, a peptide hormone and neuropeptide that signals primarily through the secretin receptor (SCTR), a class B GPCR that activates adenylyl cyclase and cAMP/PKA-dependent transcriptional programs. Secretin contributes to regulation of pancreatic bicarbonate-rich fluid secretion, biliary function, and gastrointestinal motility, and also modulates neuronal excitability and hypothalamic–pituitary signaling. Through cAMP-linked pathways and downstream ion transport processes, Sct influences epithelial fluid homeostasis and neuroendocrine communication. Altered secretin signaling has been studied in contexts of pancreatic and biliary dysfunction, metabolic and appetite regulation, and central nervous system phenotypes relevant to neurodevelopmental and gastrointestinal research.
secretin CRISPR/Cas9 KO Plasmid (m) is a pool of plasmids designed for targeted disruption of the Sct gene in mouse cell lines. Each plasmid co-expresses a unique single guide RNA (sgRNA) targeting a distinct site within the Sct together with the Streptococcus pyogenes Cas9 nuclease. The plasmids also encode GFP, allowing fluorescent identification and enrichment of successfully transfected cells by fluorescence microscopy or flow cytometry.
The multi-guide design increases the likelihood of generating insertions or deletions (indels) that disrupt the Sct open reading frame following Cas9-mediated double-strand break formation. DNA breaks introduced by the CRISPR/Cas9 system are repaired through endogenous non-homologous end joining (NHEJ) pathways, frequently resulting in frameshift mutations that abolish secretin protein expression.
This CRISPR knockout system enables efficient generation of Sct-deficient cell models for investigation of secretin signaling, functional genomics studies, cancer biology research, and evaluation of therapeutic responses in human cell lines.
