PPAR α, PPAR β, PPAR γ 와PPAR γ 2를 위한 PPAR 항체

PPARγ 항체(E-8)는 마우스, 쥐, 인간에서 유래된 PPARγ 단백질을 웨스턴 블롯(WB), 면역침전법(IP), 면역형광법(IF), 파라핀 포매 섹션(IHCP)을 이용한 면역조직화학, 효소결합 면역흡착 분석법(ELISA)을 통해 검출하는 마우스 단일클론 IgG1 kappa 경쇄 항체입니다. PPARγ(E-8) 항체는 비접합 형태와 아가로스, 피코에리틴, 플루오레세인 이소티오시아네이트, 다중 알렉사 플루오르® 접합체를 포함한 다양한 접합 형태로 제공됩니다. PPARγ 단일클론 항체(E-8)는 PPARγ1과 PPARγ2 유전자 산물을 모두 검출하는 데 권장됩니다. 핵 호르몬 수용체 슈퍼패밀리의 일원인 PPARγ는 인슐린 민감성, 지방세포 분화, 지질 대사와 관련된 유전자 발현을 조절하는 데 중요한 역할을 합니다. 레티노이드 X 수용체(RXRs)와의 이종이합체 형성은 중요한 대사 과정에 관여하는 유전자 전사를 조절하여 비만, 당뇨병, 심혈관 질환과 같은 상태의 병태 생리학에 영향을 미칩니다. PPARγ는 염증을 조절하고 면역 항상성 유지에 필수적인 대식세포의 활동에 영향을 미칩니다. PPARγ 마우스 단일클론 항체(E-8)는 대사 장애와 전사 활성을 활성화할 수 있는 프로스타노이드와 티아졸리딘디온을 포함한 다양한 리간드와의 상호작용에 대한 연구를 지원합니다. PPARγ2 유전자의 Pro12Ala 다형성은 변형된 전사 활성화 능력과 관련이 있으며, 산화된 저밀도 지단백질에 대한 면역 반응에 영향을 미쳐 제2형 당뇨병과 복부 비만과 관련될 수 있습니다.

感谢您的提问，PPAR gamma 抗体 E-8 是小鼠单克隆 IgG1 κ。二抗请选用其他种属抗小鼠。稀释比例为1:50-1:500。谢谢。 Thank you for your question, PPAR gamma antibody E-8 is a mouse monoclonal IgG1 κ. For secondary antibody, please use an anti-mouse of another species. The dilution ratio is 1:50-1:500. Thank you.

Does this product react with the human placenta?

Does HEK293 cells express this protein or could HEK293 cells be stimulated to express it?

Thank you for your question. Unfortunately we are unable to answer the expression level in HEK293 cells specifically, but this information is probably available in literature, so I would recommend looking in pubmed. We do have data in many cell lines, and we do see PPARγ expression without stimulation in most cases.

The bands we are obtaining in a WB of PAG with this antibody are around 45kDa, really close to the actin band. I am using a gel concentration of 8%. Is this band that I am obtaining PPAR gamma?? Is there any publication that can prove me it is OK?

Thank you for your question. Yes, PPARgamma has been reported at several different sizes, ranging down to 43kDa and up to 67kDa. I would suggest not using actin as a loading control when you are working with PPARgamma. GAPDH or Vinculin may be better so as to not be in the same range as your target protein.

hi, what blocking buffer that i should use with this antibody? is it better with BSA or non-fat milk?

Thank you for your question. For Western blotting with chemiluminescent detection (HRP), we recommend using 5% nonfat milk + TBS + 0.05% Tween-20 (available pre-made as, TBS Blotto A: sc-2333). For Western blotting with near-infrared (NIR) detection, we recommend using our UltraCruz® Blocking Reagent: sc-516214.
