Plectin 항체(10F6)는 마우스, 랫트, 인간에서 유래된 Plectin 단백질을 웨스턴 블롯(WB), 면역침전법(IP), 면역형광법(IF), 면역조직화학, 효소결합면역흡착분석법(ELISA)을 통해 검출하는 마우스 단일클론 IgG1 kappa 경쇄 항체입니다. 항플렉틴 항체(10F6)는 비접합형과 아가로스, 과산화효소(HRP), 피코에리트린(PE), 플루오레세인 이소티오시아네이트(FITC), 다중 알렉사 플루오르® 접합체를 포함한 다양한 접합형 형태로 제공됩니다. 플렉틴은 세포의 완전성을 유지하는 데 중요한 역할을 하는 핵심 세포 골격 단백질로서, 중간 필라멘트를 미세소관과 미세 필라멘트에 교차 결합시켜 세포질을 안정화시킵니다. 중간 필라멘트를 원형질막과 핵막에 모두 고정시키는 플렉틴의 능력은 세포 구조와 기능에 필수적입니다. 플렉틴은 라민 B와 비멘틴과 같은 주요 단백질과 상호 작용하며, 이러한 상호 작용은 다양한 단백질 키나아제에 의해 조절됩니다. 예를 들어, 단백질 키나아제 A 또는 단백질 키나아제 C에 의한 인산화는 라민 B에 대한 결합 친화성을 감소시키는 반면, 단백질 키나아제 A에 의한 인산화는 비멘틴과의 상호 작용을 향상시킵니다. 플렉틴은 p34cdc2 키나아제의 기질 역할을 하여 세포주기 조절에 관여하는 것을 강조합니다. 플렉틴의 여러 가지 대체 스플라이스 이소폼이 존재하며, 인간 플렉틴 유전자의 돌연변이는 근이영양증을 동반한 단순 표피 용해증(EBS-MD)과 관련이 있어, 구조적 및 병리학적 맥락에서 그 중요성을 강조합니다.

This is a mouse monoclonal antibody raised against purified glioma C6 cell Plectin of rat origin. The exact epitope recognized by this monoclonal antibody has not been determined. However, the following reference may be helpful, PMID: 1704007.

Is it possible to use this antibody for IF or IHC on mouse tissue samples?

Yes, this antibody comes conjugated to HRP (sc-33649 HRP), 200 µg/ml, for WB, IHC(P) and ELISA; phycoerythrin (sc-33649 PE), fluorescein (sc-33649 FITC), Alexa Fluor® 488 (sc-33649 AF488) or Alexa Fluor® 647 (sc-33649 AF647), 200 µg/ml, for IF, IHC(P) and FCM.

Hi, I am attempting to use this antibody (plectin 10F6: sc-33649) on formalin fixed,paraffin embedded archival ovarian cancer samples. Can you please give me some recommendations for antigen retrieval please, have tried microwave 15 mins low and high pH

Thank you for your question. We have not yet tested this antibody, sc-33649: Plectin (1046) using formalin-fixed paraffin-embedded tissue sections so we do not have any specific recommendations for antigen retrieval using this antibody at this time. This antibody is recommended for IF, or immunofluorescent staining of fresh or frozen tissue sections or cells fixed in acetone or methanol. In general, we recommend using a sodium citrate buffer, pH 6 using a heat treatment in a microwave for antigen retrieval.
