Phd Antibody (G-7) is a mouse monoclonal IgG2a antibody that detects phosducin in mouse, rat, and human samples through applications such as western blotting (WB), immunoprecipitation (IP), immunofluorescence (IF), and enzyme-linked immunosorbent assay (ELISA). Anti-phosducin antibody (G-7) recognizes phosducin, a crucial phototransducing protein that plays a significant role in the feedback regulation of visual phototransduction and the integration of photoreceptor metabolism. Phosducin, encoded by a gene located on chromosome 1q25-q31.1, consists of 246 amino acids and is predominantly expressed in the retina and pineal gland, with lower expression levels found in the liver, spleen, striated muscle, and brain. Within the retina, phosducin is localized specifically to the outer and inner segments of photoreceptor cells, including the synaptic and nuclear layers, where phosducin modulates the phototransduction cascade by binding to and sequestering G beta/gamma subunits of heterotrimeric G proteins. This interaction inhibits G protein-mediated signaling, preventing G alpha from reassembling with G beta/gamma and forming a functional G protein trimer, which is essential for proper signal transduction. Additionally, phosducin has been shown to slow down the internalization of G protein-coupled opioid receptors, further highlighting phosducin′s importance in cellular signaling pathways. Phosducin (G-7) antibody serves as an invaluable tool for researchers studying the intricate mechanisms of phototransduction and related signaling processes.
