Dual specificity phosphatase 2 (DUSP2), also known as PAC-1, is a MAP kinase phosphatase that dephosphorylates activated ERK1/2 and p38, thereby shaping the magnitude and duration of MAPK signaling. In human immune and hematopoietic contexts, DUSP2 contributes to stimulus-dependent control of cytokine-driven transcriptional programs, proliferation, and apoptosis by functioning as a negative feedback regulator of MAPK cascades. Its expression is often inducible in activated lymphocytes and can modulate downstream pathways linked to inflammatory signaling and stress responses. Dysregulated DUSP2 activity or expression has been associated with immune dysfunction and cancer-related signaling phenotypes, making it a useful node for mechanistic studies of MAPK pathway control.
PAC-1 CRISPR/Cas9 KO Plasmid (h) is a pool of plasmids designed for targeted disruption of the DUSP2 gene in human cell lines. Each plasmid co-expresses a unique single guide RNA (sgRNA) targeting a distinct site within the DUSP2 together with the Streptococcus pyogenes Cas9 nuclease. The plasmids also encode GFP, allowing fluorescent identification and enrichment of successfully transfected cells by fluorescence microscopy or flow cytometry.
The multi-guide design increases the likelihood of generating insertions or deletions (indels) that disrupt the DUSP2 open reading frame following Cas9-mediated double-strand break formation. DNA breaks introduced by the CRISPR/Cas9 system are repaired through endogenous non-homologous end joining (NHEJ) pathways, frequently resulting in frameshift mutations that abolish PAC-1 protein expression.
This CRISPR knockout system enables efficient generation of DUSP2-deficient cell models for investigation of PAC-1 signaling, functional genomics studies, cancer biology research, and evaluation of therapeutic responses in human cell lines.
