p-Rb Antibody (A-5) is a mouse monoclonal IgG2b kappa light chain antibody that specifically detects dually-phosphorylated forms of p-Rb at Ser 249 and Thr 252 in mouse, rat, and human samples through western blotting (WB), immunoprecipitation (IP), immunofluorescence (IF), immunohistochemistry, and enzyme-linked immunosorbent assay (ELISA). p-Rb (A-5) antibody is available in non-conjugated and various conjugated formats, including agarose, horseradish peroxidase (HRP), phycoerythrin (PE), fluorescein isothiocyanate (FITC), and multiple Alexa Fluor® conjugates. Retinoblastoma protein (Rb) regulates cell cycle progression at the G1-S transition by acting as a transcriptional repressor. Rb phosphorylation at Ser 249 and Thr 252, mediated by cyclin D-dependent kinases, inactivates Rb and enables cell cycle progression. This modification marks a shift from hypophosphorylated Rb, which actively represses cell cycle genes, to hyperphosphorylated Rb, which permits transcription of genes needed for cell division. Rb phosphorylation dynamics and regulatory mechanisms provide crucial insights into tumorigenesis pathways, especially in pediatric cancers like retinoblastoma where Rb gene mutations disrupt normal cell cycle control.
