NFκB p52 항체(C-5)는 웨스턴 블롯(WB), 면역침전법(IP), 면역형광법(IF), 유세포분석법(FCM)을 통해 쥐, 쥐, 인간에서 유래된 NFκB p52 단백질을 검출하는 마우스 단일클론 IgG2a 카파 경쇄 항체입니다. NFκB p52(C-5) 항체는 비접합 형태와 아가로스, 과산화효소, 피코에리트린, 플루오레세인 이소티오시아네이트, 다중 알렉사 플루오르® 접합체를 포함한 다양한 접합체 형태로 제공됩니다. NF-κB p52는 면역 반응, 세포 생존, 염증 조절에 중요한 역할을 담당하기 때문에, NF-κB p52는 암과 자가면역질환을 포함한 다양한 질병을 이해하는 데 중요한 표적이 되고 있습니다. NF-κB p52는 전구체인 p105의 처리를 통해 생성되며, p65와 같은 다른 NF-κB 서브유닛과 이종이합체를 형성하여 표적 유전자의 전사를 활성화합니다. I just ordered the NFkB2 antibody (C-5) sc-7386-AF647. Can you send a FCM intracellular staining protocol? Does it work with BD cyto/fix/perm and cyto/wash buffer?

Thank you for your question. You can find our flow cytometry protocol under this link: https://www.scbt.com/resources/protocols/flow-cytometry We recommend using FCM Fixation Buffer (sc-3622) and FCM Wash buffer (sc-3624) for flow cytometry.

Are there citations of this antibody being used in flow cytometry?

Hi, I am just wondering the data sheet for NFκB p52 (C-5): sc-7386 has only a single band which is not p52 but looks like p100. why this western results is different from the one on its website? thanks

Thank you for your question. The Western blot data that is currently on the data sheet is not representative of the endogenous NFKB2 protein. This is Western blot data in mouse Nfkb2 transfected 293T whole cell lysate. This transfected lysate is overepressing the full length p100 protein.

May I know what is the packaging volume does NFκB p52 Antibody (C-5) FITC (sc-7386 FITC; 200 µg/ml) come as? How many tests can be performed using flow cytometer? Thank you

Thank you for your question. We provide the FITC-conjugated primary antibody as 200 µg in 1 mL total volume. We recommend using approximately 1 µg of FITC conjugated primary antibody for each flow cytometry experiment (based on the parameters in our protocol which is provided below). The WB of the C-5 clone showed a strong band in P100, how can you use this clone for IF or IHC for NFkB2 activation, normally if P52 goes up , then P100 goes down, vice versa.

Thank you for your question. The antibody will detect p100 and/or p52, depending on what is present in the sample at the time of antibody addition. If both are present, both will be detected and you would not be able to tell them apart. I would suggest also checking your sample for the presence of one or both forms before doing your IHC or IF experiment. How can this mouse monoclonal antibody be used for double staining, if the other primary antibodies are also raised in mouse?

We recommend using directly conjugated primary antibodies. This product, sc-7386, is available conjugated to AF488, AF647, and FITC.
