MATN1 encodes matrilin-1, a cartilage-enriched extracellular matrix adaptor protein that helps organize the pericellular matrix by linking collagens and proteoglycans into supramolecular networks. Matrilin-1 contributes to chondrocyte differentiation, cartilage structural integrity, and extracellular matrix assembly during skeletogenesis and endochondral ossification. Through its modular domains and multimerization, it participates in matrix remodeling processes that influence tissue biomechanics and cell–matrix signaling. Altered MATN1 expression or matrix organization is studied in the context of cartilage degeneration and inherited or acquired skeletal and joint disorders where extracellular matrix homeostasis is disrupted.
Matrilin-1 CRISPR/Cas9 KO Plasmid (h) is a pool of plasmids designed for targeted disruption of the MATN1 gene in human cell lines. Each plasmid co-expresses a unique single guide RNA (sgRNA) targeting a distinct site within the MATN1 together with the Streptococcus pyogenes Cas9 nuclease. The plasmids also encode GFP, allowing fluorescent identification and enrichment of successfully transfected cells by fluorescence microscopy or flow cytometry.
The multi-guide design increases the likelihood of generating insertions or deletions (indels) that disrupt the MATN1 open reading frame following Cas9-mediated double-strand break formation. DNA breaks introduced by the CRISPR/Cas9 system are repaired through endogenous non-homologous end joining (NHEJ) pathways, frequently resulting in frameshift mutations that abolish Matrilin-1 protein expression.
This CRISPR knockout system enables efficient generation of MATN1-deficient cell models for investigation of Matrilin-1 signaling, functional genomics studies, cancer biology research, and evaluation of therapeutic responses in human cell lines.
