Gephyrin 항체(G-6)는 마우스, 쥐, 인간에서 유래된 Gephyrin 단백질을 웨스턴 블롯(WB), 면역침전법(IP), 면역형광법(IF), 면역조직화학, 효소결합면역흡착분석법(ELISA)을 통해 검출하는 마우스 단일클론 IgG2b 카파 경쇄 항체입니다. 항게피린 항체(G-6)는 비접합형과 아가로스, 과산화효소(HRP), 피코에리트린(PE), 플루오레세인 이소티오시아네이트(FITC), 다중 알렉사 플루오르® 접합체를 포함한 다양한 접합형 형태로 제공됩니다. 게피린은 중추신경계에서 억제성 신경전달을 유지하는 데 필수적인 억제성 글리신 수용체의 시냅스 후 국소화에 중요한 역할을 합니다. 게피린은 주로 뇌에서 발현되는데, 게피린이 시냅스 후 신경막에 국한되어 글리신 수용체의 시냅스 하 미세소관에 대한 고정을 촉진합니다. 게피린의 적절한 기능은 시냅스 안정성과 가소성에 필수적이며, 따라서 게피린은 시냅스 강도와 뇌의 흥분성 및 억제성 신호 전반의 균형을 조절하는 데 핵심적인 역할을 합니다. 쥐와 인간의 뇌 조직에서 적어도 다섯 가지의 게피린 스플라이스 변종이 확인되었으며, 각각이 신경 신호 전달에서 다양한 기능적 역할을 할 가능성이 있습니다.

I came across that some of postsynaptic proteins, including gephyrin, required milder fixation for IHC (e.g., 1-2 h 2-4% PFA fixation). If I use this antibody (G-6, sc-25311) for IHC of mouse brain sections, will it be essential to do so?

How much protein do you recommend loading on the gel for Western blot detection using Gephyrin (G-6): sc-25311?

IF: Epifluorescent analysis primary hippocampal neurons at day in vitro 15 using Gephyrin (sc-25311; 1:50) (red), GFP (green) and MAP2 (blue). Fixing in 4% PFA + 4% sucrose in PSB for 15 minutes followed 100% methanol post-fixing for 10 minutes at -20*C. WB: Analysis of Gephyrin from P30 mouse brain lysate. Mouse anti-Gephyrin (sc-25311) was diluted 1:1000 in TBS 1x containing 5% skimmed milk and 0,2% Tween-20. Anti-mouse HRP-conjugated secondary antibody was diluted 1:10000.
