Cytostatin is a cell adhesion inhibitor, inhibiting the adhesion of B16 melanoma cells to laminin and collagen type IV in a dose dependent manner but not to fibronectin. It is also an antitumor agent (antimetastatic), apoptosis inducer, and potent and selective PP2A-C (protein phosphatase 2A) inhibitor. Cytostatin interferes with the tyrosine phosphorylation of focal adhesion proteins such as focal adhesion kinase (FAK) and paxillin, which are for cell adhesion to fibronectin, a component of the ECM. This selective inhibition of PP2A by cytostatin leads to increased intracellular serine/threonine-phosphorylated proteins, modifying cell adhesion dynamics and potentially contributing to its anti-metastatic effects.
