BAP1 항체(C-4)는 마우스, 랫트, 인간에서 유래된 BAP1 단백질을 웨스턴 블롯(WB), 면역침전법(IP), 면역형광법(IF), 면역조직화학, 효소결합면역흡착분석법(ELISA)을 통해 검출하는 마우스 단일클론 IgG1 카파 경쇄 항체입니다. BAP1 항체(C-4)는 비접합형과 아가로스, 과산화효소(HRP), 피코에리트린(PE), 플루오레세인 이소티오시아네이트(FITC), 다중 알렉사 플루오르® 접합체 등 다양한 접합체 형태로 제공됩니다. BRCA1-associated protein 1로도 알려진 BAP1은 단백질 분해 조절과 세포 항상성 유지에 필수적인 유비퀴틴 가수분해 효소로서 기능합니다. 항-BAP1 항체(C-4)는 BRCA1에 의한 세포 성장 억제를 강화하여 암 예방에 기여합니다. 염색체 17q에 위치한 BRCA1 유전자의 돌연변이는 유방암과 난소암의 조기 발병률이 높은 가족의 약 45%에서 발견됩니다. BAP1은 729개의 아미노산으로 구성되어 있으며, 종양 억제에 관여하는 BARD1과 같은 다른 단백질과 상호 작용합니다. BAP1(C-4) 항체가 BRCA1에 결합하고 중요한 세포 경로에 관여하기 때문에 이 제품은 암 연구 및 치료제 개발에 유용합니다.

Which BAP-1 antibody would be best for IHC-staining in uveal melanoma (human tissue)? Is there a protocol we could use?

What codons of the BAP1 protein is this antibody detecting?

Thank you for your question. The epitope for this antibody maps within amino acids 430-729 of BAP 1 of human origin (accession#Q92560). The exact epitope has not been mapped.

What is a good control tissue to use for BAP1  immunohistochemistry optimisation?

Hi, I'm having difficulty optimizing lot H0417. I are experiencing excessive background staining. (We ran our on Ultra Benchmark, CC1 and 64minutue, option 4 at 16 minute, Antibody incubation time is 1 hr, blocker). Please advise.

Although I tried to optimize the conditions, the background was still high. Although the efficiency of antibody varies among different batches. I tried 3 different batches; only one worked but produced high background, while the other two batches did not work at all in if, despite using the same conditions.

Rated 1 out of 5 by from Inconstant staining resultsWith one batch (test sample), we obtained plausible staining results (Ventana; CC1/32min, 8' amplification, dilution 1:75/32min; nuclear loss in the tumor cells with mutation; nuclear expression in vascular endothelia and leukocytes as well as in tumors without mutation). With another batch (regularly ordered) identical staining procedures revealed strong background staining, which remained even at much higher dilutions of the antibody even in a mutated case.
