3BP2 Antibody (E-3) is a mouse monoclonal IgG1 antibody that detects 3BP2 in mouse, rat, and human samples through applications such as western blotting (WB), immunoprecipitation (IP), immunofluorescence (IF), and enzyme-linked immunosorbent assay (ELISA). 3BP2 is a crucial Syk family kinase-interacting protein (SKIP) that plays a significant role in T cell signaling, particularly in the activation and regulation of immune responses. 3BP2 is primarily located in the cytoplasm of T lymphocytes, where 3BP2 is involved in the translocation to the membrane upon T cell receptor stimulation. Membrane relocation allows for interaction with protein tyrosine kinase complexes, facilitating binding to phosphorylated Syk, LAT, and ZAP-70 proteins, which are essential for downstream signaling pathways. Furthermore, overexpression of 3BP2, particularly SH2 and proline-rich domains, can lead to the activation of key transcription factors such as NFAT and AP-1, ultimately resulting in the upregulation of the IL-2 gene promoter. This highlights 3BP2′s importance in mediating T cell activation and suggests potential as a target for therapeutic interventions in immune-related disorders.
