TOMM20 encodes Tom20, a core receptor of the translocase of the outer mitochondrial membrane (TOM) complex that recognizes N-terminal mitochondrial targeting sequences and initiates import of nuclear-encoded proteins into mitochondria. By governing mitochondrial protein biogenesis, Tom20 influences oxidative phosphorylation, mitochondrial dynamics, mitophagy, and stress signaling pathways that shape cellular metabolism and survival. Altered TOMM20 expression or mitochondrial import efficiency has been linked to mitochondrial dysfunction observed across neurodegeneration, cardiometabolic disorders, and cancer-associated metabolic remodeling. Tom20 is therefore widely used as a functional node for studying mitochondrial homeostasis, proteostasis, and organelle-to-nucleus communication.
Tom20 CRISPR/Cas9 KO Plasmid (h) is a pool of plasmids designed for targeted disruption of the TOMM20 gene in human cell lines. Each plasmid co-expresses a unique single guide RNA (sgRNA) targeting a distinct site within the TOMM20 together with the Streptococcus pyogenes Cas9 nuclease. The plasmids also encode GFP, allowing fluorescent identification and enrichment of successfully transfected cells by fluorescence microscopy or flow cytometry.
The multi-guide design increases the likelihood of generating insertions or deletions (indels) that disrupt the TOMM20 open reading frame following Cas9-mediated double-strand break formation. DNA breaks introduced by the CRISPR/Cas9 system are repaired through endogenous non-homologous end joining (NHEJ) pathways, frequently resulting in frameshift mutations that abolish Tom20 protein expression.
This CRISPR knockout system enables efficient generation of TOMM20-deficient cell models for investigation of Tom20 signaling, functional genomics studies, cancer biology research, and evaluation of therapeutic responses in human cell lines.
