NCOAT Antibody (G-12) is a mouse monoclonal IgG2b kappa light chain antibody that detects NCOAT protein of human origin by western blotting (WB), immunoprecipitation (IP), immunofluorescence (IF), immunohistochemistry with paraffin-embedded sections (IHCP), and enzyme-linked immunosorbent assay (ELISA). NCOAT (G-12) antibody is available in non-conjugated and various conjugated forms, including agarose, horseradish peroxidase (HRP), phycoerythrin (PE), fluorescein isothiocyanate (FITC), and multiple Alexa Fluor® conjugates. NCOAT protein, also known as nuclear cytoplasmic O-GlcNAcase and acetyltransferase, or MGEA5 (meningioma-expressed antigen 5), is a bifunctional enzyme that plays a crucial role in cellular regulation by acting as both a β-hexosaminidase and a histone acetyltransferase. NCOAT is predominantly expressed in the placenta, brain, and pancreas, where NCOAT catalyzes the removal of O-GlcNAc residues from glycoproteins, thereby influencing various signaling pathways and cellular processes. NCOAT′s ability to acetylate specific lysine residues on histones H3 and H4 highlights a significant role in gene expression regulation through modifications of the histone code. NCOAT enzymatic activity is optimal at a slightly acidic pH of 5.7-7, and NCOAT function can be inhibited by free N-acetylglucosamine, which underscores the importance of substrate availability in modulating activity. NCOAT is expressed as three isoforms due to alternative splicing, with isoform 1 primarily localized in the cytoplasm and isoform 3 found in the nucleus, indicating a complex regulatory mechanism that may be critical for diverse biological functions.

I am using NCOAT (G-12): sc-376429 monoclonal antibody to stain paraffin embedded tissue. Do you recommend antigen retrieval when using this antibody?
