GCAP1 Antibody (G2G4) is a mouse monoclonal IgG2a antibody that detects guanylate cyclase-activating protein 1 (GCAP1) in human and bovine samples through western blotting (WB) and immunoprecipitation (IP) applications. GCAP1 plays a crucial role in the recovery of the dark state of rod photoreceptors after exposure to light by mediating the intracellular stimulation of guanylate cyclase, which is essential for the synthesis of cyclic guanosine monophosphate (c-GMP) in photoreceptors. GCAP1 is predominantly located in the outer segments of rod and cone photoreceptors, where phototransduction occurs, highlighting GCAP1′s importance in visual signal transduction. Unlike other calcium-binding proteins in the calmodulin superfamily, the calcium-free form of GCAP1 uniquely stimulates the effector enzyme, making GCAP1 a vital component in the phototransduction cascade. Molecular cloning studies have shown that mammalian GCAPs, including GCAP1, exhibit over 90% similarity, consisting of 201 to 205 amino acids and featuring three conserved calcium-binding sites, which are critical for their function. Additionally, GCAP2, a related protein resulting from a gene duplication event, is exclusively found in the retina, further emphasizing the specialized role of GCAP proteins in visual processes.
