    SEE ALSO...

Sos 2 Antibody (G-7) is a mouse monoclonal IgG2b antibody that detects Sos 2 in human samples through various applications including western blotting (WB), immunoprecipitation (IP), immunofluorescence (IF), immunohistochemistry with paraffin-embedded sections (IHCP), and enzyme-linked immunosorbent assay (ELISA). Sos 2, a member of the guanine nucleotide exchange factor family, plays a crucial role in the activation of Ras proteins, which are pivotal in regulating cell growth, differentiation, and survival. Sos 2 facilitates the exchange of GDP for GTP on Ras, thereby promoting the active form of Ras that interacts with downstream effectors such as Raf, leading to the activation of the MAP kinase signaling pathway. This pathway is essential for transmitting signals from cell surface receptors to the nucleus, influencing gene expression and cellular responses. Sos 2 (G-7) antibody enables researchers to examine signaling networks involved in cancer and other diseases, providing detailed insights into Sos 2′s role in Ras activation and subsequent effects on cellular functions.

Can Sos 2 (G-7): sc-25350 monoclonal antibody be used to stain formalin-fixed, paraffin-embedded (FFPE) tissue sections?

    Asked by: DefinitelyNotMatt
Thank you for your question. Yes, Sos 2 (G-7): sc-25350 has been validated for use in IHC with paraffin-embedded sections. We recommend performing antigen retrieval with sodium citrate buffer (pH 6) and heat. The full protocol can be found here: https://www.scbt.com/scbt/resources/protocols/immunoperoxidase-staining

Rated 1 out of 5 by from anti-SOS2 clone G-7 did not work in a WesternI've been struggling to see a signal for SOS2 in Westerns of HeLa lysates. A coworker tested several antibodies from other vendors and chose one that just wasn't very potent. I tested 3 SCBT antibodies (clones A-8, B-6, and G-7) in a single experiment: HeLa cells vs LN229 cells, treated with an inhibitor or DMSO control and then stimulated with EGF for 5'; 40µg whole cell lysate (using NP40) per lane on a 3%-8% tris-acetate gel run at 85V for 1.2hrs, then turned up to 135V till the dye ran off the bottom. Protein transfer was to a single PVDF membrane in an iBlot2 with a custom program (created to transfer large proteins). After transfer, the blot was cut into 3 blocks. All blocks were blocked with 5% BSA in TBS for 1.5hrs, then Tween-20 was added to 0.1%, primary antibody was added at a 1:400 dilution, and the blots were incubated overnight in a cold room on a rocker. The following day, the blots were washed 4 times (5' each) with TBST, then incubated with Licor IR800 secondary antibody. Incubation was for 1.5hrs after which they were washed 4 times (5' each) and imaged using a Licor Odyssey. Only the B6 antibody showed banding. Clone G-7 produced no banding at all as seen in the attached image.
