BASP1 (Nap-22) is a membrane-associated, myristoylated protein enriched in neuronal and other excitable cells where it contributes to cortical actin organization, membrane dynamics, and phosphoinositide-dependent signaling. It modulates transcriptional programs through interactions with nuclear factors such as WT1 and participates in processes including neurite outgrowth, synaptic function, and cell differentiation. BASP1 activity has been linked to pathways governing cytoskeletal remodeling, signal transduction at the plasma membrane, and epigenetic control of gene expression. Altered BASP1 expression or regulation has been reported across multiple disease contexts, including neurodevelopmental phenotypes and cancer-associated changes in proliferation and invasion programs.
Nap-22 CRISPR/Cas9 KO Plasmid (h) is a pool of plasmids designed for targeted disruption of the BASP1 gene in human cell lines. Each plasmid co-expresses a unique single guide RNA (sgRNA) targeting a distinct site within the BASP1 together with the Streptococcus pyogenes Cas9 nuclease. The plasmids also encode GFP, allowing fluorescent identification and enrichment of successfully transfected cells by fluorescence microscopy or flow cytometry.
The multi-guide design increases the likelihood of generating insertions or deletions (indels) that disrupt the BASP1 open reading frame following Cas9-mediated double-strand break formation. DNA breaks introduced by the CRISPR/Cas9 system are repaired through endogenous non-homologous end joining (NHEJ) pathways, frequently resulting in frameshift mutations that abolish Nap-22 protein expression.
This CRISPR knockout system enables efficient generation of BASP1-deficient cell models for investigation of Nap-22 signaling, functional genomics studies, cancer biology research, and evaluation of therapeutic responses in human cell lines.
