MDM2 Antibody (HDM2-323) is a mouse monoclonal IgG2a antibody raised against amino acids 389-402 of human MDM2, suitable for applications such as western blotting (WB) and immunoprecipitation (IP), and reactive with human, mouse, and rat samples. MDM2 (Murine Double Minute 2) is an oncoprotein that serves as a crucial negative regulator of the tumor suppressor p53, playing a vital role in cell cycle control and apoptosis. MDM2 functions as an E3 ubiquitin ligase, promoting the ubiquitination and subsequent proteasomal degradation of p53, thus controlling cellular levels and activity. Post-translational modifications of MDM2, including phosphorylation, ubiquitination, sumoylation, and acetylation, are essential for modulating stability, localization, and interaction with p53 and other regulatory proteins. For instance, phosphorylation of MDM2 by kinases such as ATM, Akt, or MAPK can enhance or inhibit ubiquitination of p53, affecting the cellular response to DNA damage and stress. Ubiquitination of MDM2 can lead to degradation, creating an autoregulatory feedback loop that fine-tunes p53 activity. These post-translational modifications regulate the MDM2-p53 pathway, ensuring proper cell cycle progression and apoptosis. Dysregulation of MDM2′s modifications can result in impaired p53 function, leading to uncontrolled cell proliferation and tumorigenesis.
