Col4a2 encodes the α2 chain of type IV collagen (COL4A2), a core structural component of basement membranes that forms a heterotrimeric network with other collagen IV chains to support tissue architecture. This extracellular matrix scaffold regulates cell adhesion, migration, and polarity and shapes signaling crosstalk with integrins and growth factor pathways involved in vascular stability and organ development. Altered COL4A2 function is linked to basement membrane fragility and small-vessel pathology, making it relevant to studies of cerebrovascular integrity, ocular development, and kidney filtration barriers. In mouse models, Col4a2 provides a tractable entry point for dissecting matrix-driven mechanisms that influence endothelial and epithelial behavior.
COL4A2 CRISPR/Cas9 KO Plasmid (m) is a pool of plasmids designed for targeted disruption of the Col4a2 gene in mouse cell lines. Each plasmid co-expresses a unique single guide RNA (sgRNA) targeting a distinct site within the Col4a2 together with the Streptococcus pyogenes Cas9 nuclease. The plasmids also encode GFP, allowing fluorescent identification and enrichment of successfully transfected cells by fluorescence microscopy or flow cytometry.
The multi-guide design increases the likelihood of generating insertions or deletions (indels) that disrupt the Col4a2 open reading frame following Cas9-mediated double-strand break formation. DNA breaks introduced by the CRISPR/Cas9 system are repaired through endogenous non-homologous end joining (NHEJ) pathways, frequently resulting in frameshift mutations that abolish COL4A2 protein expression.
This CRISPR knockout system enables efficient generation of Col4a2-deficient cell models for investigation of COL4A2 signaling, functional genomics studies, cancer biology research, and evaluation of therapeutic responses in human cell lines.
