CATSPER1 encodes CatSper, a sperm-specific ion channel subunit required for calcium influx that drives hyperactivated motility and supports capacitation-associated signaling. CatSper channel activity integrates changes in membrane potential and intracellular Ca2+ dynamics to regulate flagellar beating patterns and downstream phosphorylation events essential for fertilization. Disruption of CATSPER1 is linked to impaired sperm function and is associated with male infertility phenotypes, making it a key target for mechanistic studies of sperm motility control. As a component of a multi-subunit channel complex, CatSper1 also serves as an entry point to investigate assembly, trafficking, and regulation of sperm ion channels.
CatSper CRISPR/Cas9 KO Plasmid (h) is a pool of plasmids designed for targeted disruption of the CATSPER1 gene in human cell lines. Each plasmid co-expresses a unique single guide RNA (sgRNA) targeting a distinct site within the CATSPER1 together with the Streptococcus pyogenes Cas9 nuclease. The plasmids also encode GFP, allowing fluorescent identification and enrichment of successfully transfected cells by fluorescence microscopy or flow cytometry.
The multi-guide design increases the likelihood of generating insertions or deletions (indels) that disrupt the CATSPER1 open reading frame following Cas9-mediated double-strand break formation. DNA breaks introduced by the CRISPR/Cas9 system are repaired through endogenous non-homologous end joining (NHEJ) pathways, frequently resulting in frameshift mutations that abolish CatSper protein expression.
This CRISPR knockout system enables efficient generation of CATSPER1-deficient cell models for investigation of CatSper signaling, functional genomics studies, cancer biology research, and evaluation of therapeutic responses in human cell lines.
