BrdU Antibody (IIB5) is a mouse monoclonal IgG1 kappa light chain antibody that detects BrdU protein in non-species specific applications through immunofluorescence (IF), immunohistochemistry with paraffin-embedded sections (IHCP), and flow cytometry (FCM). Anti-BrdU antibody (IIB5) is available in non-conjugated and various conjugated forms, including agarose, horseradish peroxidase (HRP), phycoerythrin (PE), fluorescein isothiocyanate (FITC), and multiple Alexa Fluor® conjugates. The halogenated pyrimidine analog bromodeoxyuridine (BrdU) plays a crucial role in DNA synthesis, incorporating into newly synthesized DNA strands during the S-phase of the cell cycle, making BrdU an invaluable tool for estimating the fraction of cells actively undergoing DNA replication. This incorporation allows researchers to quantify DNA synthesis and study cellular processes such as sister chromatid exchange and nascent DNA isolation. BrdU (IIB5) antibody binds specifically to exposed BrdU in single-stranded DNA following hydrochloric acid denaturation or nuclease digestion, which is essential for accurate detection. For optimal results, protease antigen recovery is recommended for tissues or cells fixed with crosslinking agents like formalin, although this may affect BrdU immunodetection specificity. BrdU (IIB5) monoclonal antibody shows cytoplasmic staining, providing clear visualization in immunohistochemistry and immunocytochemistry applications, thereby facilitating deeper understanding of cellular proliferation and DNA synthesis dynamics.
