Arsenite methyltransferase (AS3MT) is a key enzyme involved in the biotransformation of inorganic arsenic, a common environmental contaminant that poses serious health risks to humans. AS3MT is present in a variety of organisms, including humans, and is especially important for detoxifying arsenic in the body.AS3MT is primarily found in the liver, where the majority of arsenic methylation takes place. The enzyme uses S-adenosylmethionine (SAM) as the methyl donor in the methylation process. AS3MT adds a methyl group to inorganic arsenic to form monomethylarsonic acid (MMA), and then another methyl group to MMA to form dimethylarsinic acid (DMA). This methylation process is believed to facilitate the excretion of arsenic from the body, as the methylated forms of arsenic (MMA and DMA) are more readily excreted in the urine than the inorganic forms. However, the methylation process also creates intermediate forms of arsenic that are more toxic than the inorganic forms. For example, MMA(III), the trivalent form of MMA, is considered more toxic than inorganic arsenic.
Arsenic is typically ingested in its inorganic forms, arsenite (As(III)) or arsenate (As(V)). These are highly toxic and carcinogenic, but the body can reduce their toxicity through methylation. This is where AS3MT comes in. AS3MT catalyzes the transfer of a methyl group from S-adenosylmethionine (SAM) to arsenic, converting it first to monomethylarsonic acid (MMA) and then to dimethylarsinic acid (DMA), both of which are less toxic and more readily excreted than the inorganic forms. The efficiency of this methylation process can vary greatly among individuals due to genetic differences in the AS3MT gene, which can affect the activity of the enzyme. This can influence an individual's susceptibility to arsenic toxicity. Some polymorphisms in the AS3MT gene have been associated with an increased risk of arsenic-related health effects, such as skin lesions and bladder cancer.
