BFP or Blue Fluorescent Protein is a derivative of Green Fluorescent Protein (GFP).. BFP was one of the first color variants of GFP to be engineered and is characterized by its blue fluorescence.BFP works similarly to GFP. It consists of a sequence of amino acids that forms a barrel-like structure, inside of which is a chromophore, the part of the molecule responsible for its fluorescent properties. When BFP absorbs light of a specific wavelength, its chromophore is excited to a higher energy state. When it returns to its ground state, it emits light of a longer wavelength, which is seen as blue fluorescence.
BFP has been used as a marker in various biological research applications, including studying protein localization, gene expression, and protein-protein interactions in living cells. It has been particularly useful in multicolor labeling strategies, where it can be used in combination with other fluorescent proteins to simultaneously track multiple biological processes.However, BFP is less commonly used than other fluorescent proteins due to its relatively low brightness and stability, and its need for a UV light source for optimal excitation, which can be damaging to living cells. Improved versions of BFP, such as Azurite and mTagBFP, have been developed to overcome some of these limitations.
