Glyceraldehyde-3-phosphate dehydrogenase (GAPDH), also called uracil DNA glycosylase, catalyzes the reversible oxidative phosphorylation of glyceraldehyde-3-phosphate in the presence of inorganic phosphate and nicotinamide adenine dinucleotide (NAD), an important energy-yielding step in carbohydrate metabolism. While GAPDH has long been recognized as playing an integral role in glycolysis, additional functions of GAPDH include acting as a uricil DNA glycosylase, activating transcription, binding RNA and involvement in nuclear RNA export, DNA replication and DNA repair. Expression of GAPDH is upregulated in liver, lung and prostate cancers. GAPDH translocates to the nucleus during apoptosis. GAPDH complexes with neuronal proteins implicated in human neuro-degenerative disorders including the β-Amyloid precursor, Huntingtin and other triplet repeat neuronal disorder proteins.

Does this antibody show cross-reactivity with E. coli GAPDH?

Thank you for your inquiry. The immunogen used to raise our GAPDH antibody sc-166574 shows low homology with the sequence for E.coli GAPDH, so we do not predict it will detect this protein and have no data to support reactivity.

Rated 3 de 5 de par discreto40ug of protein were loaded in AnyKD precasted gel Electrophoresis was performed in TGS buffer at 100V Immunoblot was performed with Protran Nitrocellulose Membrane at 400mA for 90’ Membranes were incubated 1h in BSA 5% in TBS + Tween 0.1% (TBS-T) GAPDH antibody was diluted 1:1000 in TBS-T and membranes incubated o/n at 4°C Membranes were washed three times with TBS-T for 10’ Secondary antibody anti-mouse HRP conjugated (Bio-Rad 170-6516) was diluted 1:20000 in TBS-T and added to membranes for 1h at RT Signal was detected with ECL (GE GE RPN2209) in the dark room Exposition time 1’ 3’ 15’ In our experimental conditions the antibody worked well and the signal was detected at expected molecular weight
