    Overview

    NKX2-3 encodes the homeobox transcription factor Nkx-2.3, a sequence-specific DNA-binding regulator involved in developmental patterning and cell fate decisions. It is best known for roles in endoderm-derived tissues, including intestinal and splenic organogenesis, where it helps coordinate transcriptional programs that shape tissue architecture. Nkx-2.3 influences gene networks controlling epithelial differentiation, regional identity, and immune-associated stromal functions, linking it to pathways that govern mucosal homeostasis. Genetic and expression alterations in NKX2-3 have been associated with inflammatory bowel disease susceptibility and related dysregulation of gut immune interactions, making it relevant for mechanistic studies of intestinal inflammation and barrier biology.

    Nkx-2.3 CRISPR/Cas9 KO Plasmid (h) is a pool of plasmids designed for targeted disruption of the NKX2-3 gene in human cell lines. Each plasmid co-expresses a unique single guide RNA (sgRNA) targeting a distinct site within the NKX2-3 together with the Streptococcus pyogenes Cas9 nuclease. The plasmids also encode GFP, allowing fluorescent identification and enrichment of successfully transfected cells by fluorescence microscopy or flow cytometry.

    The multi-guide design increases the likelihood of generating insertions or deletions (indels) that disrupt the NKX2-3 open reading frame following Cas9-mediated double-strand break formation. DNA breaks introduced by the CRISPR/Cas9 system are repaired through endogenous non-homologous end joining (NHEJ) pathways, frequently resulting in frameshift mutations that abolish Nkx-2.3 protein expression.

    This CRISPR knockout system enables efficient generation of NKX2-3-deficient cell models for investigation of Nkx-2.3 signaling, functional genomics studies, cancer biology research, and evaluation of therapeutic responses in human cell lines.
