Mouse Hamp encodes hepcidin, a liver-derived peptide hormone that serves as a central regulator of systemic iron homeostasis. Hepcidin binds the iron exporter ferroportin (SLC40A1) to trigger its internalization and degradation, thereby limiting intestinal iron absorption and controlling iron release from macrophages and hepatocytes. Its transcription is controlled by inflammatory signaling such as IL-6/STAT3 and by iron-sensing pathways including BMP/SMAD, integrating innate immunity with iron metabolism. Dysregulation of the hepcidin–ferroportin axis is implicated in iron-loading and iron-restricted states and is widely studied in models of anemia of inflammation, hemochromatosis-like phenotypes, and infection-associated nutritional immunity.
hepcidin CRISPR/Cas9 KO Plasmid (m) is a pool of plasmids designed for targeted disruption of the Hamp gene in mouse cell lines. Each plasmid co-expresses a unique single guide RNA (sgRNA) targeting a distinct site within the Hamp together with the Streptococcus pyogenes Cas9 nuclease. The plasmids also encode GFP, allowing fluorescent identification and enrichment of successfully transfected cells by fluorescence microscopy or flow cytometry.
The multi-guide design increases the likelihood of generating insertions or deletions (indels) that disrupt the Hamp open reading frame following Cas9-mediated double-strand break formation. DNA breaks introduced by the CRISPR/Cas9 system are repaired through endogenous non-homologous end joining (NHEJ) pathways, frequently resulting in frameshift mutations that abolish hepcidin protein expression.
This CRISPR knockout system enables efficient generation of Hamp-deficient cell models for investigation of hepcidin signaling, functional genomics studies, cancer biology research, and evaluation of therapeutic responses in human cell lines.
