FGF19 encodes fibroblast growth factor 19 (FGF-19), an endocrine FGF that signals primarily through FGFR4 in conjunction with the co-receptor β-Klotho to regulate metabolic homeostasis. In hepatocytes and enterocytes, FGF-19 participates in bile acid feedback control by suppressing CYP7A1 and integrating with FXR-driven transcriptional programs that influence lipid and glucose metabolism. Downstream signaling engages MAPK/ERK and related kinase cascades, linking FGF-19 to cell growth, differentiation, and tissue-specific metabolic adaptation. Dysregulated FGF19 expression or signaling has been associated with altered bile acid metabolism and metabolic disease phenotypes, and it is frequently studied in the context of aberrant growth signaling in cancer biology.
FGF-19 CRISPR/Cas9 KO Plasmid (h) is a pool of plasmids designed for targeted disruption of the FGF19 gene in human cell lines. Each plasmid co-expresses a unique single guide RNA (sgRNA) targeting a distinct site within the FGF19 together with the Streptococcus pyogenes Cas9 nuclease. The plasmids also encode GFP, allowing fluorescent identification and enrichment of successfully transfected cells by fluorescence microscopy or flow cytometry.
The multi-guide design increases the likelihood of generating insertions or deletions (indels) that disrupt the FGF19 open reading frame following Cas9-mediated double-strand break formation. DNA breaks introduced by the CRISPR/Cas9 system are repaired through endogenous non-homologous end joining (NHEJ) pathways, frequently resulting in frameshift mutations that abolish FGF-19 protein expression.
