DNAI1 encodes dynein intermediate chain 1, an axonemal dynein component required for assembly and force transmission within outer dynein arms that power motile cilia and flagella beating. By coupling ATP-driven dynein motor activity to microtubule doublets, Dynein IC1, axonemal contributes to coordinated ciliary waveform generation and mucociliary clearance processes. DNAI1 function intersects with pathways governing cilia biogenesis, axonemal organization, and intraflagellar transport-dependent maintenance of motile cilia structure. Disruption of DNAI1 is associated with primary ciliary dyskinesia, linking altered axonemal dynein integrity to impaired ciliary motility and downstream developmental and respiratory phenotypes.
Dynein IC1, axonemal CRISPR/Cas9 KO Plasmid (h) is a pool of plasmids designed for targeted disruption of the DNAI1 gene in human cell lines. Each plasmid co-expresses a unique single guide RNA (sgRNA) targeting a distinct site within the DNAI1 together with the Streptococcus pyogenes Cas9 nuclease. The plasmids also encode GFP, allowing fluorescent identification and enrichment of successfully transfected cells by fluorescence microscopy or flow cytometry.
The multi-guide design increases the likelihood of generating insertions or deletions (indels) that disrupt the DNAI1 open reading frame following Cas9-mediated double-strand break formation. DNA breaks introduced by the CRISPR/Cas9 system are repaired through endogenous non-homologous end joining (NHEJ) pathways, frequently resulting in frameshift mutations that abolish Dynein IC1, axonemal protein expression.
This CRISPR knockout system enables efficient generation of DNAI1-deficient cell models for investigation of Dynein IC1, axonemal signaling, functional genomics studies, cancer biology research, and evaluation of therapeutic responses in human cell lines.
