Adrenomedullin (ADM) is a multifunctional peptide hormone that plays a pivotal role in several physiological processes, including vasodilation, cell growth, and immune response modulation. It was initially discovered in human pheochromocytoma tissue and is widely expressed in various organs such as the lungs, kidneys, and cardiovascular system. The primary ADM signaling pathway involves its binding to a G protein-coupled receptor (GPCR) complex consisting of the calcitonin receptor-like receptor (CLR) and receptor activity-modifying proteins (RAMPs). This receptor binding activates adenylate cyclase, leading to an increase in cyclic AMP (cAMP) levels within the cell. Elevated cAMP activates protein kinase A (PKA), which then phosphorylates various downstream targets to mediate the diverse physiological effects of ADM. Besides the canonical CLR/RAMP receptor pathway, ADM also interacts with other receptor types, such as L1 cell adhesion molecule, to engage in more complex signaling networks.
