VAChT Activators

The chemical class that indirectly activates VAChT is diverse, encompassing a range of neurotransmitters, ion channel modulators, and ions. These chemicals mainly function by modulating the activity of the cholinergic neurons and the production, release, or degradation of acetylcholine, which the VAChT protein transports into synaptic vesicles. The overall activity of VAChT is dependent on the rate of acetylcholine synthesis and breakdown, as well as the excitability of the neurons. Thus, any chemical that influences these factors can indirectly alter VAChT activity.

The neurotransmitters dopamine, GABA, and glutamate, for instance, can influence acetylcholine synthesis and release, which in turn can modulate VAChT activity. Similarly, ions, such as calcium, sodium, and potassium, are essential for maintaining the membrane potential, the function of the SLC family of transporters, and the release of acetylcholine, indirectly affecting VAChT activity. The GABA receptor antagonist bicuculline and the AMPA/kainate receptor antagonist CNQX, by modulating the activity of GABAergic and glutamatergic neurons respectively, can also indirectly influence VAChT. Lastly, the sodium channel modulators veratridine and tetrodotoxin can alter cellular excitability and therefore indirectly modulate VAChT activity.
