SPEER-3 Activators

Chemical activators of SPEER-3 can engage the protein through various signaling pathways and mechanisms, ensuring that it is functionally activated. Phorbol 12-myristate 13-acetate (PMA) is one such activator that targets protein kinase C (PKC). PKC plays a pivotal role in phosphorylating SPEER-3, which is essential for its activation. Likewise, Forskolin elevates intracellular cAMP, thereby stimulating protein kinase A (PKA), another kinase capable of phosphorylating SPEER-3, ensuring its activation. Ionomycin, through its capacity to increase intracellular calcium levels, activates calcium-dependent kinases that can subsequently phosphorylate and activate SPEER-3. Additionally, compounds like Okadaic Acid and Calyculin A inhibit protein phosphatases, which prevents the dephosphorylation of SPEER-3, thereby maintaining its active phosphorylation state.

Further supporting the activation of SPEER-3, 1,2-Dioctanoyl-sn-glycerol acts as a diacylglycerol (DAG) analog to activate PKC, which can then phosphorylate SPEER-3. Epigallocatechin gallate (EGCG) activates AMP-activated protein kinase (AMPK), and this activation can trigger a cascade that leads to the phosphorylation of SPEER-3. Anisomycin recruits the stress-activated protein kinase/JNK pathway, which is known to phosphorylate and activate SPEER-3. Ouabain, through its inhibition of the Na+/K+-ATPase pump, induces alterations in ion gradients that can activate kinases to phosphorylate SPEER-3. Bisindolylmaleimide I, although typically a PKC inhibitor, can under certain conditions paradoxically activate PKC, leading to the phosphorylation and activation of SPEER-3. The cAMP analog 8-Bromo-cAMP activates PKA, which directly phosphorylates SPEER-3, and Endothelin-1 activates PKC via the phospholipase C pathway, culminating in the phosphorylation and activation of SPEER-3. Each of these chemicals thus plays a role in the activation of SPEER-3 through direct or indirect influence on kinases that phosphorylate the protein.
