Chemical activators of Smok1 can trigger its functional activation through various intracellular signaling mechanisms. Phorbol 12-myristate 13-acetate (PMA), a well-known activator of Protein Kinase C (PKC), can lead to the activation of Smok1 by promoting PKC to phosphorylate Smok1. Similarly, Forskolin raises intracellular cAMP levels, which activate Protein Kinase A (PKA), another kinase that can phosphorylate and thus activate Smok1. Ionomycin, by increasing intracellular calcium levels, can activate calcium-dependent kinases which then phosphorylate Smok1, leading to its activation. Another route of activation is through the action of Epidermal Growth Factor (EGF), which binds to its receptor to initiate the MAPK/ERK pathway; this cascade can result in the activation of Smok1 through phosphorylation by involved kinases.
Additionally, certain chemicals can sustain the active state of Smok1 by inhibiting the dephosphorylation process. Okadaic Acid and Calyculin A inhibit protein phosphatases, which normally dephosphorylate proteins, thereby maintaining Smok1 in a phosphorylated and active state. Anisomycin activates stress-activated protein kinases that can phosphorylate Smok1. Dibutyryl-cAMP (db-cAMP), a cAMP analog, activates PKA, which then can activate Smok1 by phosphorylation. A23187 acts as a calcium ionophore that elevates intracellular calcium and activates kinases that phosphorylate Smok1. Sodium Orthovanadate functions by inhibiting protein tyrosine phosphatases, preserving the phosphorylated active state of Smok1. Bisindolylmaleimide I, known for its inhibitory action on PKC, can indirectly influence other kinases that phosphorylate Smok1. Lastly, Thapsigargin, by inhibiting the SERCA pump, raises intracellular calcium, which can activate kinases that phosphorylate and maintain Smok1 in an active state. Each of these chemicals, through these specific biochemical pathways, can lead to the functional activation of Smok1.
