MAP3K3, also known as Mitogen-Activated Protein Kinase Kinase Kinase 3 or MEKK3, is a protein kinase that plays a crucial role in the regulation of the MAPK (Mitogen-Activated Protein Kinase) signaling pathway. The MAP3K3 signaling pathway is initiated when extracellular ligands bind to their corresponding cell surface receptors, leading to the activation of upstream signaling molecules, such as small GTPases (Ras, Rho, etc.), which in turn activate MAP3K3. Activated MAP3K3 then phosphorylates and activates its downstream target, MAP2K (Mitogen-Activated Protein Kinase Kinase), also known as MEK. MAP2K subsequently phosphorylates and activates MAPK, which includes three main subfamilies: ERK (Extracellular Signal-Regulated Kinase), JNK (c-Jun N-terminal Kinase), and p38 MAPK. These activated MAPKs then translocate to the nucleus or other cellular compartments, where they phosphorylate and modulate the activity of specific transcription factors and other signaling proteins, leading to the expression of target genes and various cellular responses.
