LKLF Activators are a collection of chemicals that can either directly or indirectly elicit the activation or upregulation of LKLF (KLF4), a transcription factor critical in numerous cellular pathways, including differentiation and proliferation. These activators engage a variety of intracellular pathways that culminate in the enhanced expression or activity of LKLF. For instance, Retinoic Acid triggers LKLF upregulation through retinoic acid receptors (RARs), while Wnt-3a operates via the canonical Wnt/β-catenin pathway. Forskolin, on the other hand, increases intracellular levels of cAMP, which in turn facilitates LKLF transcription. PPARγ agonists, such as Rosiglitazone, upregulate LKLF through activation of PPARγ, while 5-Aza-2'-deoxycytidine promotes LKLF expression by demethylating its gene promoter.

For example, Rapamycin inhibits mTOR, which then indirectly leads to an increase in LKLF levels. Vitamin D3 (Cholecalciferol) enhances LKLF transcription through VDR activation, while Dexamethasone influences LKLF through the glucocorticoid receptor. Hydrogen Peroxide, a cellular stress inducer, is also able to upregulate LKLF.
