Chemical inhibitors of LIN-44 can disrupt the signaling pathways where this protein is involved by targeting various steps of the Wnt cascade. D4476 acts by inhibiting casein kinase 1, a kinase implicated in the phosphorylation of Wnt proteins, which is essential for their function. The inhibition of CK1 by D4476 can therefore interfere with the post-translational modifications that LIN-44 requires for its activity. Similarly, IWP-2 and LGK-974 impede the activity of porcupine, an enzyme required for the palmitoylation of Wnt proteins. Since LIN-44 is a member of the Wnt family, the blockade of porcupine by these inhibitors prevents the necessary palmitoylation of LIN-44, thus blocking its ability to engage in signaling.
Further down the signaling pathway, XAV-939 and JW55 inhibit the function of tankyrase, which leads to the stabilization of Axin and the subsequent promotion of β-catenin degradation. LIN-44 signaling relies on the availability of β-catenin to activate downstream transcriptional programs, and these inhibitors hamper the pathway by reducing β-catenin levels. KY02111 contributes to this process by promoting the degradation of β-catenin through enhancement of GSK-3β activity, which forms part of the β-catenin destruction complex. Additionally, CCT251545 disrupts Wnt signaling by inhibiting TCF4, preventing the β-catenin/TCF4 complex formation necessary for LIN-44 to exert its effects. PRI-724 and ICG-001 both act by inhibiting the interaction between β-catenin and CBP, a coactivator required for the transcription of Wnt target genes. By disrupting the formation of the β-catenin/CBP complex, these inhibitors impede the transcriptional activation usually mediated by LIN-44. CGP049090 and FH535 also target β-catenin-mediated transcription, but through the inhibition of the TCF/β-catenin complex and β-catenin/TCF mediated transcription respectively, leading to a similar downregulation of LIN-44 signaling activity. WIKI4, like JW55 and XAV-939, acts upon tankyrase, further emphasizing the critical role of β-catenin regulation in controlling LIN-44 function in the cell.
