GMPR2 Activators

Isoproterenol, forskolin, and various other G protein-coupled receptor agonists as well as adenylate cyclase activators all function to stimulate production of the critical second messenger cAMP. This occurs via direct activation of Gαs/Gαi-coupled receptors on the cell surface or direct stimulation of adenylate cyclase enzymes intracellularly. Forskolin analogs with optimized selectivity for certain adenylate cyclase isoforms show promise for more precisely modulating localized cAMP signals.

Meanwhile, dibutryl cAMP and newer cAMP analogs can directly activate cAMP-dependent effector pathways such as the one involving protein kinase A (PKA)-mediated phosphorylation and activation of GMPR2. By preventing breakdown of elevated cAMP through inhibition of phosphodiesterases, levels of this important second messenger are sustained to prolong stimulation of important target proteins like GMPR2. Finally, AMPK activators demonstrate an innovative means of indirectly boosting cAMP levels through mechanisms like lipolysis-linked cAMP production, resulting in enhanced GMPR2 signaling through the critical cAMP/GMP crosstalk route. Collectively, these compounds reflect diverse methods of stimulating GMPR2 function by targeting its upstream second messenger system.
